PATHOGENESIS OF LOCAL ISOLATE OF RABBIT HAEMORRHAGIC DISEASE VIRUS (RHDV) IN BALADY RABBITS AND CELL CULTURE

Abstract

The Balady rabbif) were I/N experimentally inFected with RHDV to study the pathogenesis of The virus. The virus induced 100% morbidity and mortality. The virus antigen(s) were detected in Liver, spleen, lung, heart and kidney by HA, AGPT AND FAT. The virus amounts were inCreased gradually after infection and reached 13 Log, HA titer after 48 hours in liver tissues folLowed by spleen and lung 9 log,, heart tissues 8 Log, and finally kidney tissues 6 log, HA titer. The virus take about 5 days to express its cytoPathic effect (CPE) on primary rabbit kidney cells In the first passage and decreased to 2 days in the Fourth passage in which infectivity titer of the viRus increased 2.8 logo TCIDs0/ml than he first Passage. In case of isolation on Vero cells, the viRus take about 7 days to appear its CPE in the first Passage and decreased to 3 days in the fourth pasSage. The infectivity titer of the virus increased 3.5 logo TCID50/ml in the fourth passage than in The first passage. The haemagglutinin was loss Completely after the third passage in both cell culTure used. This study reflected that the preparation Of RHDV vaccine using cell culture is not preFerred due to the losses of HA antigen during Propagation.
